Anastrozole is as a non-steroidal aromatase inhibitor (AI) used to decrease estrogen levels. It acts by blocking the enzyme aromatase, subsequently blocking the production of estrogen.
Anastrol is indicated for the first-line treatment of postmenopausal women with hormone receptor-positive or hormone receptor unknown locally advanced or metastatic breast cancer.
For males; indicated for control of gynecomastia and water retention from concurrent treatment with testosterone or other aromatizing anabolic steroids.
Mechanism of Action
The growth of many cancers of the breast are stimulated or maintained by estrogens. In postmenopausal women, estrogens are mainly derived from the action of the aromatase enzyme, which converts adrenal androgens (primarily androstenedione and testosterone) to estrone and estradiol. The suppression of estrogen biosynthesis in peripheral tissues and in the cancer tissue itself can therefore be achieved by specifically inhibiting the aromatase enzyme. Anastrozole is a selective non-steroidal aromatase inhibitor. It significantly lowers serum estradiol concentrations and has no detectable effect on formation of adrenal corticosteroids or aldosterone. A daily dose of 1mg has been shown to reduce serum estradiol levels by greater than 80%.
May cause or increase progress of osteoporosis.
May cause hot flashes or thinning of the hair.
Negative impact on serum lipids.
Chest pain, shortness of breath, swelling of feet or lower legs.
Estrogen-containing drugs should not be used with Anastrol as they may diminish its pharmacological action.
Pregnancy, or planning to become pregnant, or are breast-feeding.
Allergies to anastrazole, similar medicines, or similar substances.
Liver disease, elevated liver enzymes, or high cholesterol.
Decreases in bone mineral density may occur. Consider bone mineral density monitoring.
In women with pre-existing ischemic heart disease, an increased incidence of ischemic cardiovascular events occurred with anastrozole use compared to tamoxifen use. Consider risks and benefits.
Serum lipids including total cholesterol, HDL, and LDL.
Serum estradiol levels.
Bone mineral density monitoring due to risk of decreased bone mineral density.
DOSAGE AND ADMINISTRATION
Women: 1mg oral tablet once daily or as directed by a physician.
Men: 0.25 to 1mg taken orally every other day or as directed by physician.
1 mg film coated tablets : 50 tablets in 5 blisters.
Protect from light. Store at 15 - 25oC.
